MOTS-c is a 16-amino-acid peptide encoded within mitochondrial DNA and naturally produced by the body. Research has shown that it functions as a metabolic signalling peptide (often referred to as a mitokine), helping coordinate communication between mitochondria and the cell nucleus during periods of metabolic stress. Experimental studies indicate that MOTS-c influences glucose and fatty acid metabolism, particularly within skeletal muscle, and activates the AMPK pathway—a key regulator of cellular energy balance. Because of these properties, it has attracted scientific interest as an exercise mimetic, with research exploring how it may influence metabolic efficiency, insulin signalling and age-related metabolic changes. Natural MOTS-c production has been reported to decline with age, contributing to ongoing investigations into its biological significance. While research continues, MOTS-c is not approved as a therapeutic medicine and is prohibited by the World Anti-Doping Agency (WADA) for competitive athletes.
